Lee Sun Kyun made his debut in 2001 as Brad in the musical The Rocky Horror Show. He starred in many popular musicals, including Grease (2003), Indian Love Song (2005), and Love, Love, Love (2013). He made his television debut in the 2002 MBC Korean drama Lovers, playing a very small supporting role. He played supporting roles in many popular Korean dramas and TV series, such as Thousand Years of Love (2003), Chahandangshin Love Crazy (2003), Transparent (2004), Pure Love (2005), Love Crazy (2005), Taereung National Village (2005), and Fugitive Lee Doo-yong (2006). Lee Sun Kyun rose to fame with the 2007 medical Korean drama White Tower.

On December 27, 2023, Lee Sun-kyun was found dead in a parked car in Wolyong Park in Seoul, South Korea. According to his family, he left home earlier that day and left a suicide note.

- In high school, he also liked music and advertising and wanted to be a radio program producer.
- Lee Sun-kyun was a member of the lighting team of the university drama club. During a drama performance, one of his seniors ran away from home and Sun-kyun was chosen as a replacement. While performing on stage, Sun-kyun developed a sense of belonging to the stage and decided to become an actor.
