Polanco wanted to be an actress since she was a child, but she was always afraid to go to auditions because of her weight. After graduation, she started working in healthcare with the idea of becoming a nurse. She was working in hospital administration at Montefiore Medical Center in the Bronx while studying nursing when she decided to explore acting again and enrolled in an acting workshop. Her first acting roles were in the television series Unforgettable and NYC 22.
Polanco was cast as Dayanara “Daya” Diaz, an inmate at a women’s prison, in the Netflix series Orange Is the New Black in 2012. She starred in the 2013 indie film Gimme Shelter. In June 2014, it was revealed that she had been promoted from a recurring character in series regular for the show’s third season, which premiered in June 2015. She starred in the comedies The Cobbler (2014) and Joy (2015) before moving on to The Perfect Match (2016).
