Maithili has been interested in acting since she was a child. She used to perform street plays in the society during festivals. Whenever there was a performance at school or college, she was the first to attend. Her passion for acting grew and she began performing in theaters. She first came to prominence in 2006 in the Marathi film Aai Mala Maaf Kar.

On March 26, 2016, Maithili Jawkar filed an FIR against Mumbai Bharatiya Janata Party (BJYM) president Ganesh Pandey. She accused him of sexual harassment during BJYM’s three-day executive committee meeting. Maithili said Ganesh invited her to his room and made lewd remarks to her. Although Ganesh resigned as BJYM president, he refuted the claims and said they were part of a conspiracy against him.
