In 2012, Priyanka married Tamil director-turned-actor Lawrence Ram in a traditional Hindu wedding at the Attukal Bhagavathy temple in Thiruvananthapuram, Kerala. However, their marriage did not last long and Priyanka filed for divorce in 2015 and they later separated. She revealed in an interview that the real reason she broke up with her ex-husband was that he spread her pornographic photos through social media and banned her from acting. Her son Mukund Ram is fondly called “Appu” by his family. After the couple decided to separate, Lawrence Ram filed a lawsuit seeking custody of his son, but Priyanka won custody of her son and Mukund lives with her .

She made her debut with Vasanthabalan’s 2006 Tamil film Veyil. Priyanka’s first Malayalam film was Kichamani MBA starring Suresh Gopi, which was released in 2007. In 2008, she made her acting debut in the Kannada film Zindagi, her only film in the Kannada film industry. In 2012, Priyanka temporarily retired from the film industry after her marriage. She returned to the Malayalam film industry with Potasbomb (2013).

In 2006, Priyanka was selected as Veyil, one of the heroines in Vasanthabalan; she played the role of ‘Thangam’ in the film. The film received positive reviews and was a box office success. Veyil was screened at the 2007 Cannes International Film Festival in the “Tous Les Cinemas Du Monde” section; it was the first Tamil film to be screened at the Festival. She received widespread appreciation for her performance as ‘Zahira’ in Chandran TV’s Vilapangalkkappuram (2008); She received positive reviews and won her first award for her role; Priyanka starred in Suresh The film Bhoomi Malayalam (2009) starring Suresh Gopi. In 2021, she played the role of Meera, a woman suffering from schizophrenia, in Abilash Purushothaman’s bilingual TV series Aa Mukham. The film was released in Malayalam and Tamil; Priyanka is the only actor to appear in the entire film. “Aa Mukham” becomes the second feature film from Kerala to feature a single female character. She has appeared in many movies including: Samastha Keralam PO (2009), Ividam Swargamanu (2009), Pottas Bomb (2013), Jalam (2016), Home (2021), The 12th Man (2022), Varal (2022) and Kaduva (2022).

Priyanka Nair acted in several television serials during her college years; as she had to juggle her studies, she became a part-time serial actress. She revealed in an interview that although she acted in several TV series during college, she did not take acting seriously or consider acting as a career plan. She wanted to become a lecturer. Priyanka played the role of Vinduja in “Oomakkuyil” (2003) and a ghost in “Akashadoothu”. (2003).
