Makarande was born on March 6, 1966 in Dahanu, Maharashtra, India. He currently resides in Vile Parle, Mumbai, Maharashtra. He became interested in writing at an early age and began writing plays in college. As of 2018, he has created more than 90 plays in his theater career, including short plays and full-length plays. For his outstanding performance, he won the South Indian International Film Award for Best Negative Actor twice in 2012 for the movies “Dandupalya” (Kannada) and “No. 66 Madhura Bus” (Malayalam) .

Makarand Deshpande studied at BPM High School, Mumbai and graduated from Narsee Monjee College of Commerce and Economics, Mumbai. While in college, he began writing plays for intercollegiate competitions. He started his acting career in 1988 playing the role of Baba in the Bollywood movie “Qayamat Se Qayamat Tak”. Apart from Bollywood, Makarande has also acted in many other language films such as “Dandupalya” (Kannada, 2012), “Jalsa” (Telugu, 2008), “No. 66 Madhura Bus” (Malayalam, 2012), Rita (Marathi, 2009), etc.
In 1990, he started working as a theater artist at Prithvi Theater and has created numerous plays. Later, he met actor Kay Kay Menon and co-founded Ansh Theater Company in 1993. Subsequently, he made his debut with the feature-length drama “Dreamman”. Makarand Deshpande has written, directed and produced many famous plays in his theatres, such as Sir Sir Sarla (2006), Karodo Mein Ek (2008), Miss Beautiful (2009), “Joke” (2009), “Dev Vanar”, “Vasant Ka Teera Youvan”, etc.
He has also written and directed films such as Danav (2003), Hanan (2004), Shahrukh Bola Khoobsurat Hai Tu (2010), Sona Spa (2013) and Saturday Sunday (2014).
In 2011, he served as one of the judges of the Marathi reality show Marathi Paul Padate Pudhe.
In 2013, Makarande participated in the sports reality TV show Celebrity Cricket League (CCL) Season 3 as a player for the Veer Marathi team.
