Yashomati Thakur entered politics as a member of the Indian National Congress (INC). In 2004, she contested the Maharashtra Assembly elections for the first time from Teosa (also known as Tivsa) constituency in Amravati district, but lost; Bharatiya Janata Party candidate Sahebrao Thatte won the seat. From 2004 to 2009, she served as the general secretary of the Maharashtra State Youth Congress. At the same time, she also served as a member of the State Women’s Commission. Later, she joined Rahul Gandhi’s Youth Brigade and was responsible for the states of Gujarat, Daman and Diu, Madhya Pradesh and Chhattisgarh. She has served as Secretary of the Maharashtra State Congress Committee, Secretary of the All-India Congress Committee, and Chairman of the Maharashtra State Congress Committee. She contested and won the Teosa Assembly seat in the 2009 Maharashtra Legislative Assembly elections, defeating Shiv Sena candidate Sanjay Band by 26,130 votes. She was re-elected as Teosa MP in 2014 and 2019. On December 30, 2019, she was sworn in as Cabinet Minister for Women and Child Development in the Maha Vikas Aghadi government led by Uddhav Thackeray; she held this position till June 29, 2022. In 2018, Congress appointed Yasomati as Karnataka party secretary. She was appointed as the President of Vidhansabha Talika by Nana Patole on December 16, 2019, to attend the winter session of Maharashtra Assembly in Vidarbha, Nagpur. As an MLA in the Maharashtra government, she served as the Guardian Minister of Amravati district (2020-2022).

2012 police assault
Yashomati Thakur and three others, including her driver, were sentenced to three months in prison by an Amravati court for allegedly assaulting a policeman in 2012 in Amravati district. She has since appealed to the High Court against the conviction, seeking a suspended sentence in the form of interim relief. The incident took place on March 24, 2012, in Chulabhatti area, which comes under the jurisdiction of Rajapeth police station in Amravati. Yashomati Thakur, her driver Sagar Suresh Khandekar and party workers Sharad Kashirav Jawanjal and Raju Kisan Ingle allegedly assaulted traffic police officer Ulhas Raurale when he stopped her vehicle from entering the single carriageway. In 2020, the court sentenced Thakur to three months’ rigorous imprisonment and fined him Rs 10 lakh in the eight-year-old case. 15,500. She was fined equal amounts by the District Court and the District Court.

- Yashomati Thakur contested against her sister Sanyogita Naik Nimbalkar in the 2014 Maharashtra Legislative Assembly elections. contested the election as an independent candidate with the support of local organization Prahar. Nimbalkar led Thakur’s campaign in the 2009 assembly elections and contributed to her victory. Addressing the media, Nimbalkar expressed her disappointment with Thakur, saying:
I campaigned for my sister during the 2009 elections and thought she would bring development. But she didn’t do anything, so I decided to take on her. “
